Christelle Hureau présentera la conférence intitulée : "Fundamental insights on the Cu and Zn-driven deleterious effects in Alzheimer & therapeutic approaches to counterbalance them"

Abstract

Alzheimer's disease (AD) is characterized by the exracellular deposits made of supramolecular assemblies of the amyloid-β (Aβs) peptides, in which up to mM level of metal ions, mainly copper and zinc has been found. Two events related to the interaction of Aβs with metal ions are involved in the etiology of AD: the production of reactive oxygen species (ROS) by Cu(Aβ) species and the assembly of the Aβs leading to various kind of aggragates, including amyloids.

During the talk, I will show e brief overview on our contribution to the elucidation of the mechanism of Cu(Aβ)-induced ROS production and on the therapeutic approaches we have set up to counteract it. Then, I will focus on our last peptide assembly data. They include the study of metal-altered co-assembly of two different Aβ peptides, with the aim to go towards more biologically relevant situations. Last, I will describe another therapeutic option that relies on the modulation on the Aβ assembly towards less toxic aggregates and illustrate it with selected examples.
