Romé Voulhoux présentera une conférence intitulée "Structure-function exploration of dynamic processes in bacterial type 2 secretion".

Abstract

Bacteria have developed sophisticated nanomachines to ensure the efficient and specific secretion of multiple extracellular factors, which are essential for their survival in often hostile environments. Among these, the type 2 secretion system (T2SS) is specialized in the transport of complex exoproteins requiring intracellular folding. The current understanding of the T2SS secretion process suggests a sequential and dynamic mechanism involving the successive interaction of the secreted effector with the secretin and endopilus components of the machinery, according to an atypical conformational recognition mode that remains unknown.

During this seminar, I will discuss how structure-function approaches involving cryo-EM and AlphaFold, combined with protein-protein interaction tools as simple as cysteine cross-linking, have enabled us to better understand several dynamic aspects of this singular secretory nanomachine. I will therefore discuss secretin ”breathing,” endopilus assembly, and, of course, substrate recognition and transport.
